Brief description

The ZEISS Elyra 7 with Lattice SIM can be used for fluorescence super-resolution microscopy. Experiments for both structured illumination microscopy (SIM) and single-molecule localization microscopy (SMLM) can be performed.

In SIM mode a lattice pattern is applied for sample illumination. Data processing is required to get a super-resolved image. Depending on the imaging algorithm lateral resolution can be improved down to ~120 nm (classical SIM processing) and further down to ~60 nm (SIM2 processing), axial resolution can be improved down to ~300 nm (classical SIM processing) and ~200 nm (SIM2 processing). Processing algorithms are incorporated in the processing section of the ZEN Blue software.

SMLM can be perfomed in TIRF or HiLo mode and resolution can be improved down to the range of 10–20 nm.

On top the Elyra 7 can be used as a stand alone TIRF setup and can as well be used in SIM Apotome mode with lower objective magnifications for fast enhanced optical sectioning with increased contrast and resolution.

For all mentioned options single or dual camera mode can be used. The system is equipped with two Hamamatsu ORCA-Fusion BT sCMOS cameras connected via the Duolink camera adapter.

The microscope is equipped with full incubation enabling 37 °C and 5 % CO2 for live cell imaging conditions.
